Cinoxacin is synthesised through a multi-step process beginning with 4,5-methylenedioxy-2-nitroacetophenone, which is hydrogenated over a platinum catalyst to yield 2-amino-4,5-methylenedioxyacetophenone. This intermediate undergoes diazotization, triggering spontaneous heterocyclization to form 4-hydroxy-6,7-methylenedioxycinnoline. The cinnoline compound is then brominated using molecular bromine and potassium acetate, producing 3-bromo-4-hydroxy-6,7-methylenedioxycinnoline. A cyanation reaction with copper(I) cyanide in dimethylformamide replaces the bromine with a cyano group, forming 3-cyano-4-hydroxy-6,7-methylenedioxycinnoline. This compound is then alkylated at the nitrogen atom using ethyl iodide and sodium hydride, followed by acidic hydrolysis of the cyano group to a carboxylic acid, yielding the final product: cinoxacin. | |||
